After completion of the mission "Freak Show" with an "Excellent" rating, the player unlocks Perry as a playable character in Showdown Mode. He can also become playable if the player buys the Monocle from the General Store after "Freak Show".
Professor Perry was the inventor of Perry's Miracle Elixir, selling his tonic to the public. He later became a member of O'Leary's Traveling Circus and Carnival Show. One day, the tents that Preston O'Leary and his performers were sleeping in just outside of Tarnation caught fire, killing them all (presumably caused by Perry himself). The law were unable to investigate this thoroughly due to a lack of manpower, so Professor Perry stepped up and became the new leader of the circus.
Perry renamed the circus into "Professor Perry's Potions & Peculiarities", he began hiring freaks, criminals, and other deranged folk. Wherever their circus would go, crime would spike in the area. This caused him and his gang to become extremely feared throughout the land.
Events of Red Dead Revolver
Prof. Perry is the final boss of the mission "Freak Show".
After Jack Swift killed all the other major members of Perry's circus (excluding Atlas Jones), Perry arrived to fight him himself. Perry would constantly vanish and re-appear in different areas, and when he would get low on health, he would drink his elixir to heal himself. After his elixir became empty, Jack Swift was able to kill Professor Perry, bringing an end to him and his circus.
Perry is listed in Sheriff Bartlett's Journal under the Prof. Perry's Peculiarities section. His journal entry occupies pages 102-103 and you unlock it by buying the Monocle for $666 in the General Store during the 2nd Brimstone free-roam section (after Chapter 7).

Trivia
Jack and Perry have an animosity towards each other, probably due to the fact that Jack tried to leave the circus. Perry also loved to taunt Jack Swift's British heritage.
Perry shares some similarities to DC Universe's Joker.
In Showdown Mode, he lacks his teleporting abilities and immunity to toxic otherwise he would have appeared too powerful.
Similar to Mr. Black, Perry might harm his own men on occasion when teleporting if they stand too close to him.
His recoloured models in Showdown Mode have him wearing either black, brown or yellow coat.
